Born on September 13, 1966, in Melbourne, Victoria, Australia, Mandylor is the son of Greek immigrants. Growing up, he was an athletic young man, playing for Heidelberg United FC Megas Alexandros in their 1988 State League championship team. He also played five National League games in 1989, scoring one goal. In addition to his football career, Mandylor was also an avid boxer, winning 14 fights with 4 losses.
Mandylor's passion for acting eventually led him to emigrate to the United States, where he began his acting career. He made his breakthrough with the role of Nick Portokalos in My Big Fat Greek Wedding, a role that would become his most iconic and beloved. The film's success led to the creation of a spinoff TV series, My Big Fat Greek Life, and two sequels, My Big Fat Greek Wedding 2 and My Big Fat Greek Wedding 3.
Mandylor has had a diverse range of roles throughout his career, including a guest appearance on Relic Hunter alongside Tia Carrere. He played Louis Malone in the first season of Martial Law alongside Sammo Hung. One of his most memorable roles was as Joey's twin in the popular sitcom Friends episode, "The One With Unagi". Ironically, Mandylor had originally auditioned for the role of Joey, but was beat out by Matt LeBlanc.
He has also appeared twice on CSI: Miami, once as a robber-killer in the third season and again in a different role in the eighth season. Mandylor has had the opportunity to work alongside his brother, Costas, in several projects, including the Charmed episode "Saving Private Leo", the crime thriller Sinners and Saints, and the short film My Brother Cicero, which Louis wrote and produced.
