Born on August 23, 1949, in Fort Wayne, Indiana, Long was raised in a Presbyterian family and developed a passion for performing at a young age. She was an active member of her high school speech team and won the National Forensic League's National Championship in Original Oratory in 1967. After studying drama at Northwestern University, Long pursued a career in acting and modeling, landing her first job as a meal plan checker at the university.
Long's breakthrough came when she began performing in local commercials for Homemakers furniture store in the Chicago area. She then joined The Second City comedy troupe, where she honed her skills as a writer, producer, and co-host of the television program Sorting It Out on WMAQ-TV. Her work on the show earned her three Regional Emmys.
Long's most notable role, of course, is that of Diane Chambers on Cheers, a character that earned her five Emmy nominations and two Golden Globe Awards. She reprised her role in three episodes of the spin-off Frasier, for which she received an additional guest star Emmy nomination.
In addition to her success on television, Long has appeared in several films, including Night Shift (1982), Irreconcilable Differences (1984), The Money Pit (1986), Outrageous Fortune (1987), Hello Again (1987), Troop Beverly Hills (1989), The Brady Bunch Movie (1995), A Very Brady Sequel (1996), and Dr. T & the Women (2000).

In her personal life, Long has been married twice and has a daughter, Juliana Thames Long.
