Born on May 8, 1977, in Athens, Greece, Papaloukas began his professional career in 1995 with his local club Ampelokipoi. He then transferred to Dafni in 1997 and later to Panionios in 1999, where his outstanding performances earned him a move to EuroLeague powerhouse Olympiacos in 2001. With Olympiacos, Papaloukas won his first title, the Greek Cup, in 2002.
In 2003, Papaloukas made a pivotal move to CSKA Moscow, where he would go on to rewrite the history books.
