In 2021, Penisini started training with the Parramatta Eels' first-grade team, and on March 4, 2021, he was upgraded to the club's 30-man squad, signing with the Eels until the end of 2022. Later that year, he resigned with the club for a further season until 2023. Penisini made his NRL debut for the Parramatta Eels against the Canberra Raiders in round 19, 2021, and scored his first try in the NRL during a 32-16 victory over the North Queensland side in round 23.
