His father, Celso Dantas Sales, was a judge in the Court of Appeals of the State of Rio Grande do Norte. From a young age, Sales was drawn to the priesthood, and he began his humanistic studies as a teenager.
In 1936, he entered the minor seminary at Natal, where he spent a year before moving on to the major seminary at Fortaleza. At the major seminary, Sales prepared for the priesthood from 1937 to 1943. He was ordained to the priesthood on November 21, 1943, and spent the following decade in pastoral work in the Archdiocese of Natal.
In 1954, Pope Pius XII appointed Sales to the episcopate, naming him titular bishop of Thibica and auxiliary bishop of Natal. Sales was consecrated a bishop on August 15, 1954. He attended all the sessions of the Second Vatican Council between 1962 and 1965, and on October 29, 1968, he was appointed Metropolitan Archbishop of São Salvador da Bahia, becoming ex officio the Primate of Brazil.
On April 28, 1969, Pope Paul VI created Archbishop Sales a Cardinal of the Holy Roman Church. On April 30, 1969, Cardinal Sales received his red biretta and title of San Gregorio VII. He served as archbishop of São Sebastião do Rio de Janeiro until his resignation was accepted in 2001, when he had already passed the maximum age for voting in a papal conclave.
