After her parents' divorce at the age of two, Tripplehorn grew up in Tulsa and graduated from Edison High School in 1981. She attended the University of Tulsa for a semester before enrolling in the Juilliard School's Drama Division, where she was part of Group 19 (1986-1990), alongside fellow actress Laura Linney.
Tripplehorn began her career on stage, appearing Off-Broadway in John Patrick Shanley's The Big Funk in 1990. She also costarred with Val Kilmer in a 1993 production of John Ford's 1630s play Tis Pity She's a Whore.
