Niarchos attended the Nardin Academy grammar school in Buffalo and later studied in the best private school in Athens. He went on to study law at the University of Athens, after which he started working for his maternal uncles in the Koumantaros family's grain business.
During World War II, Niarchos served as a naval officer, and part of the trade fleet he had developed with his uncle was destroyed. He used the insurance settlement of about two million dollars to build a new fleet, marking the beginning of his shipping empire. In 1952, he founded Niarchos Ltd., an international shipping company that would eventually operate more than 80 tankers worldwide.
Niarchos is famously known for his rivalry with Aristotle Onassis, another Greek shipping magnate. The two titans of the industry engaged in a bitter competition, with Niarchos commissioning the construction of massive oil supertankers to gain an edge in the market. His most notable asset was the yacht Atlantis, which he later gifted to King Fahd of Saudi Arabia and is now known as Issham al Baher.

He was a multiple-time leading owner and topped the French breed list, earning him a reputation as a master breeder.
