Born on July 28, 1867, in Steubenville, Ohio, Perrine was the son of a Methodist minister and a descendant of Daniel Perrin, a Huguenot. Growing up with a passion for photography and astronomy, Perrine moved to Alameda, California, in 1886 and worked as a bookkeeper at Armour Co. in San Francisco.
In 1893, Perrine joined the Lick Observatory in California, where he worked alongside prominent astronomers, honing his skills and making significant observations. During his tenure, Perrine was awarded the Lalande Prize and gold medal by the Paris Academy of Sciences in 1897 for his outstanding contributions to astronomy.
In 1909, Perrine became the Director of the Argentine National Observatory in Cordoba, Argentina, a position he held until 1936. Under his leadership, the observatory made two attempts to measure the bending of starlight near the Sun during solar eclipses, a phenomenon predicted by Einstein's theory of relativity. Although the attempts were thwarted by unfavorable weather conditions, Perrine's efforts marked the first attempts to prove the theory through astronomical observations.
