Rutherford was born on November 3, 1749, in Edinburgh, Scotland, to Anne Mackay and Professor John Rutherford. He began his educational journey at Mundells School, West Bow, and later pursued medicine under the tutelage of William Cullen and Joseph Black at the University of Edinburgh. Rutherford graduated with a doctorate in medicine (MD) in 1772.
In 1775, Rutherford practiced medicine in Edinburgh, where he became a prominent figure in the scientific community. In 1782, he co-founded the Harveian Society of Edinburgh and served as its president in 1787. Additionally, he was a joint founder of the Royal Society of Edinburgh in 1783 and a member of the Aesculapian Club in 1784.
Rutherford's most notable achievement, however, was the isolation of nitrogen in 1772. While working under Joseph Black, he discovered that a candle would not burn in carbon dioxide, leading him to explore the properties of air. Through his experiments, Rutherford successfully isolated nitrogen, paving the way for further research into atmospheric gases.
Rutherford held various prestigious positions throughout his career, including Professor of Botany at the University of Edinburgh and the 5th Regius Keeper of the Royal Botanic Garden Edinburgh from 1786 to 1819. He also served as President of the Royal College of Physicians of Edinburgh from 1796 to 1798.
Rutherford married Harriet Mitchelson of Middleton in 1786. His notable family connection is his uncle-nephew relationship with the celebrated novelist Sir Walter Scott.
