Charalambis was born on September 22, 1862, in Kalavryta, Greece. He graduated from the Hellenic Military Academy and was commissioned as a 2nd Lieutenant of Cavalry on July 25, 1884. He quickly rose through the ranks, becoming a lieutenant in 1888, captain in 1895, and major in 1908. During the Balkan Wars of 1912-1913, he served as Chief of Staff of the 1st Infantry Division in Macedonia and the 6th Infantry Division in Epirus.
In 1922, following the defeat of Greece against Turkey in Anatolia, Charalambis played a crucial role in the political crisis that ensued. A military revolt led by Venizelist officers demanded the resignation of King Constantine I and Prime Minister Nikolaos Triantafyllakos. Charalambis was appointed Minister of Military Affairs in the new government, with Alexandros Zaimis as Prime Minister. When Zaimis was out of the country, Charalambis briefly served as interim Prime Minister of Greece on September 29, 1922.
